- American Idealism: Uncle Sam's powers are proportionate to the United States' faith in the ideals of liberty and freedom.
- Possession: The American Spirit will merge with a dying patriot to manifest physically. Uncle Sam will usurp all memories of the host. The possession is permanent until that Sam chooses voluntarily to leave the body.
- Immortality: Uncle Sam's appearance, though occasionally different, remains relatively the same in every year. He always appears as a middle-aged though able-bodied man with white hair and a white beard.
- Interdimensional Teleportation: He is also shown to be able to transport himself and others to a pocket of an alternate dimension called The Heartland. Pre-Crisis, he was also able to travel between Earth-Two and Earth-X at will, though he lost this ability when fatigued.
- Invulnerability: While he has survived direct explosions, Uncle Sam is able to be knocked unconscious if caught off guard.
- Invisibility:
- Limited Clairvoyance: While on Earth-X in 1941, Uncle Sam received a vision of the attack on Pearl Harbor occurring on Earth-Two. The vision came unbidden, and was not something Sam could control.
- Super-Leaping:
- Size Alteration: Uncle Sam can increase his size at will. He once increased his size to the point where he could grab two flying nuclear warheads and keep them from reaching their destinations.

- Superhuman Strength: He once tore the hull of a Japanese battleship and sank it with his bare hands.
- Super-Breath:
- Swimming: Uncle Sam was able to swim to the bottom of the ocean off the coast of Long Island. While early on in his adventures, it seemed like he could breathe underwater and even talk, later escapades revealed that he could only hold his breath for a limited time.
- Diving:
- Danger Sense: Uncle Sam once confided to Buddy that he receives "a direct appeal or a sign" when he's needed, but even he does not fully understand it.
- Bio-Fission: In great American civil conflicts, Sam can split in two entities, each one supporting one of the sides. During the American Civil War, Sam had been split in two as Johnny Reb and Billy Yank. It's unknown if he can split in more duplicates.
- Sam can detect the whereabouts of patriotic objects, as the American Constitution, the Declaration of Independence and possibly others.
- Dimensional Travel: Uncle Sam was able to return to The Clouds, home of the Spirit People, and back when America lost faith in him.
- Fourth Wall Awareness: Uncle Sam was able to best King Killer by jumping between panels.

- Disguise: Uncle Sam is adept at impersonating various characters, and once disguised himself as Adolf Hitler.
- Fighting Mastery: Buddy claimed that Uncle Sam was a "champion at any kind of fighting." Whether this meant he had the ability to flawlessly match peak performance in any form of fighting or that he had trained extensively to learn virtually every style of fighting is unknown.
- Fencing: He fought and disarmed a would-be murderer in a foreign country during a duel.

- Marksmanship: Uncle Sam showed his prowess in marksmanship several times from throwing mines from land to sink ships at sea, or rocks that ricochet to eliminate several enemies at once.
- Multilingualism:
- Tracking: Uncle Sam taught Buddy how to traverse forest terrain and track, skill he himself learned from Native Americans.

- Uncle Sam's Airplane: Uncle Sam's airplane was a 2-engine bomber, with 1930s-style U.S. military markings, and a large white numeral "4" on the fuselage. It was uniquely sized and shaped.
- Uncle Sam's Car: Uncle Sam's car was a 1920s-looking open-top jalopy that never seemed to run out of gas.
- Bumble-Boat: A yellow submarine invented by Buddy, capable of travels over 120 mph at sea. It is equipped with torpedoes powerful enough to sink specialized battleships in one hit. They were later mass produced for the United States Navy with an insignia of Uncle Sam and Buddy's face.

- American Patriotism: Uncle Sam's powers are directly linked to the American people, their fighting spirit, and faith in their country and Uncle Sam. A lack of faith result in Uncle Sam becoming weaker and weaker to the point of powerlessness.
- Bondage: Uncle Sam regularly performs great feats of strength, but an inventor only known as "Doc" once created and bound Uncle Sam with with unbreakable string. Only after Buddy was in danger could Uncle Sam break it.
- Magic: When Uncle Sam was hit with a Mayan magic-infused poison, he was left in a severely weakened state.
- Spiritual Dependency: Uncle Sam's powers are grounded by the strength of the American consciousness. When the overarching spirit of America falters, Sam's strength and stamina decline. During the American Civil War, the country's faith was sundered completely, and Sam was split into two composite forms, Johnny Reb and Billy Yank.
